Giemsa stain solution is intended for the detection and confirmation of blood protozoa and microfilariae. Thick smears are better for detecting parasites because larger volumes are examined. However, thin smears are better for identifying individual organisms. The staining solution is a solution of giemsa stain in methanol and glycerin. Giemsa stain itself is a compound stain formed from polychromed methylene blue and eosin. The working solution is prepared just before use by diluting the stock solution with a phosphate buffer. Buffers with a pH between 6.8 and 7.2 are usually chosen, with 6.8 or 7.0 being the most common. The resulting stain solution must be used immediately.
